Sky gel for Tissue culture

Sky Gel can be used for Tissue Culture also following the hints provided herewith.

1. Skygel basically retains its water absorbent strength even when subjected to autoclave either in granule form or mixed with water.

2. But one of the major hurdles is the exposure to light, as UV light disintegrates the Skygel (Thats why in horticulture we recommend burying of Skygel particles under the soil).

3. If there is a feasibility, the UV light 100% prevention could be ideal, but practically it may be very difficult.

4. In such cased the following methods could be employed to have a good growth in Skygel for Tissue culture:

STEP: 1:

The sides of the tissue culture bottle or tupper ware should be first painted black to prevent exposure to light from the side portion.

STEP: 2:

To prevent exposure to light from above, a thin aluminium foil can be spreaded on the surface of the Skygel and required number(s) of slit openings may be made to have plant(s) as shown in the illustration on the right.

This is better achieved when tupper wares are used which have square or rectangular shaped bottom and same size caps. But in bottles with round caps which are smaller than the base this is difficult.

STEP: 3:

Another option instead of No. 2 is to cover the upper portion of the Skygel with fine charcoal particles which will prevent light exposure from above. Even mixing of charcoal particles could also be made.
